Pancreatitis is a complex, progressively destructive inflammatory disorder. Alcohol was long thought to be the primary causative agent, but genetic contributions have been of interest since the discovery that rare PRSS1, CFTR and SPINK1 variants were associated with pancreatitis risk. We now report two associations at genome-wide significance identified and replicated at PRSS1-PRSS2 and X-linked CLDN2 through a two-stage genome-wide study. The PRSS1 variant likely affects disease susceptibility by altering expression of the primary trypsinogen gene. The CLDN2 risk allele is (...) associated with atypical localization of claudin-2 in pancreatic acinar cells. The homozygous CLDN2 genotype confers the greatest risk, and its alleles interact with alcohol consumption to amplify risk. The use of human fetal tissue for scientific research has enormous potential but is subject to government legislation. In the United Kingdom the Polkinghorne Committee's guidelines were accepted by the Department of Health in 1990. These guidelines set out to protect women undergoing termination of pregnancy from exploitation but in so doing may significantly restrict potential research. Although the committee took evidence from a wide variety of experts they did not seek the views of the general public. We asked 108 (...) women about to have a therapeutic abortion; 167 women who had had a pregnancy terminated in the past, and 419 women who had never had an abortion, their views on research using human fetal tissue. Regardless of their past experiences the women were overwhelmingly in favour of research using fetal tissue (94 per cent). They made little distinction between basic research and research with obvious clinical relevance and supported the concept of using transplanted fetal tissue for the treatment of adult disease such as Parkinsonism. Women about to undergo an abortion were significantly more likely (p < 0.001) to approve of all types of research including that aimed at improving methods of abortion and research using live fetuses in utero. In this article, we present results from a literature review of intrinsic, instrumental, and relational values of nature conducted for the Intergovernmental Science-Policy Platform on Biodiversity and Ecosystem Services, as part of the Methodological Assessment of the Diverse Values and Valuations of Nature. We identify the most frequently recurring meanings in the heterogeneous use of different value types and their association with worldviews and other key concepts. From frequent uses, we determine a core meaning for each value type, which is (...) sufficiently inclusive to serve as an umbrella over different understandings in the literature and specific enough to help highlight its difference from the other types of values. Finally, we discuss convergences, overlapping areas, and fuzzy boundaries between different value types to facilitate dialogue, reduce misunderstandings, and improve the methods for valuation of nature's contributions to people, including ecosystem services, to inform policy and direct future research. © 2023 The Author(s). Practitioners and advocates of community food security (CFS) envision food systems that are decentralized, environmentally-sound over a long time-frame, supportive of collective rather than only individual needs, effective in assuring equitable food access, and created by democratic decision-making. These themes are loosely connected in literature about CFS, with no logical linkages among them. Clear articulation in a theoretical framework is needed for CFS to be effective as a guide for policy and action. CFS theory should delimit the level of analysis (...) (i.e., what are the boundaries of “community”); show how CFS relates to individual, household, and national food security and explain emergent properties, which are important at the community level of analysis; point to the best indicators of CFS or its lack; clarify the determinants of CFS; and clarify the stages of movement toward CFS. This theoretical base would allow researchers to develop valid and reliable measures, and allow practitioners to weigh alternative options to create strategic plans. A theoretical base also would help establish common ground with potential partners by making the connections to anti-hunger work, sustainable agriculture, and community development clear. (shrink)

The focus of this study was to analyze the content, distribution, and comparative genome relationships of 996 chromosome bin-mapped expressed sequence tags accounting for 2266 restriction fragments on the homoeologous group 3 chromosomes of hexaploid wheat. Of these loci, 634, 884, and 748 were mapped on chromosomes 3A, 3B, and 3D, respectively. The individual chromosome bin maps revealed bins with a high density of mapped ESTs in the distal region and bins of low density in the proximal region of the (...) chromosome arms, with the exception of 3DS and 3DL. These distributions were more localized on the higher-resolution group 3 consensus map with intermediate regions of high-mapped-EST density on both chromosome arms. Gene ontology classification of mapped ESTs was not significantly different for homoeologous group 3 chromosomes compared to the other groups. A combined analysis of the individual bin maps using 537 of the mapped ESTs revealed rearrangements between the group 3 chromosomes. Approximately 232 of the consensus mapped ESTs matched sequences on rice chromosome 1 and revealed large- and small-scale differences in gene order. Of the group 3 mapped EST unigenes ∼21 and 32% matched the Arabidopsis coding regions and proteins, respectively, but no chromosome-level gene order conservation was detected. (shrink)

A total of 1918 loci, detected by the hybridization of 938 expressed sequence tag unigenes from 26 Triticeae cDNA libraries, were mapped to wheat homoeologous group 4 chromosomes using a set of deletion, ditelosomic, and nulli-tetrasomic lines. The 1918 EST loci were not distributed uniformly among the three group 4 chromosomes; 41, 28, and 31% mapped to chromosomes 4A, 4B, and 4D, respectively. This pattern is in contrast to the cumulative results of EST mapping in all homoeologous groups, as reported (...) elsewhere, that found the highest proportion of loci mapped to the B genome. Sixty-five percent of these 1918 loci mapped to the long arms of homoeologous group 4 chromosomes, while 35% mapped to the short arms. The distal regions of chromosome arms showed higher numbers of loci than the proximal regions, with the exception of 4DL. This study confirmed the complex structure of chromosome 4A that contains two reciprocal translocations and two inversions, previously identified. An additional inversion in the centromeric region of 4A was revealed. A consensus map for homoeologous group 4 was developed from 119 ESTs unique to group 4. Forty-nine percent of these ESTs were found to be homoologous to sequences on rice chromosome 3, 12% had matches with sequences on other rice chromosomes, and 39% had no matches with rice sequences at all. Limited homology was found between wheat ESTs on homoeologous group 4 and the Arabidopsis genome. Forty-two percent of the homoeologous group 4 ESTs could be classified into functional categories on the basis of blastX searches against all protein databases. (shrink)

The objectives of this study were to develop a high-density chromosome bin map of homoeologous group 7 in hexaploid wheat, to identify gene distribution in these chromosomes, and to perform comparative studies of wheat with rice and barley. We mapped 2148 loci from 919 EST clones onto group 7 chromosomes of wheat. In the majority of cases the numbers of loci were significantly lower in the centromeric regions and tended to increase in the distal regions. The level of duplicated loci (...) in this group was 24% with most of these loci being localized toward the distal regions. One hundred nineteen EST probes that hybridized to three fragments and mapped to the three group 7 chromosomes were designated landmark probes and were used to construct a consensus homoeologous group 7 map. An additional 49 probes that mapped to 7AS, 7DS, and the ancestral translocated segment involving 7BS also were designated landmarks. Landmark probe orders and comparative maps of wheat, rice, and barley were produced on the basis of corresponding rice BAC/PAC and genetic markers that mapped on chromosomes 6 and 8 of rice. Identification of landmark ESTs and development of consensus maps may provide a framework of conserved coding regions predating the evolution of wheat genomes. (shrink)

The complex hexaploid wheat genome offers many challenges for genomics research. Expressed sequence tags facilitate the analysis of gene-coding regions and provide a rich source of molecular markers for mapping and comparison with model organisms. The objectives of this study were to construct a high-density EST chromosome bin map of wheat homoeologous group 2 chromosomes to determine the distribution of ESTs, construct a consensus map of group 2 ESTs, investigate synteny, examine patterns of duplication, and assess the colinearity with rice (...) of ESTs assigned to the group 2 consensus bin map. A total of 2600 loci generated from 1110 ESTs were mapped to group 2 chromosomes by Southern hybridization onto wheat aneuploid chromosome and deletion stocks. A consensus map was constructed of 552 ESTs mapping to more than one group 2 chromosome. Regions of high gene density in distal bins and low gene density in proximal bins were found. Two interstitial gene-rich islands flanked by relatively gene-poor regions on both the short and long arms and having good synteny with rice were discovered. The map locations of two ESTs indicated the possible presence of a small pericentric inversion on chromosome 2B. Wheat chromosome group 2 was shown to share syntenous blocks with rice chromosomes 4 and 7. (shrink)

This report describes the rationale, approaches, organization, and resource development leading to a large-scale deletion bin map of the hexaploid wheat genome. Accompanying reports in this issue detail results from chromosome bin-mapping of expressed sequence tags representing genes onto the seven homoeologous chromosome groups and a global analysis of the entire mapped wheat EST data set. Among the resources developed were the first extensive public wheat EST collection. Described are protocols for sequencing, sequence processing, EST nomenclature, and the assembly of (...) ESTs into contigs. These contigs plus singletons were used for selection of distinct sequence motif unigenes. Selected ESTs were rearrayed, validated by 5′ and 3′ sequencing, and amplified for probing a series of wheat aneuploid and deletion stocks. Images and data for all Southern hybridizations were deposited in databases and were used by the coordinators for each of the seven homoeologous chromosome groups to validate the mapping results. Results from this project have established the foundation for future developments in wheat genomics. (shrink)

The thymus is a unique primary lymphoid organ that supports the production of self‐tolerant T‐cells essential for adaptive immunity. Intrathymic microenvironments are microanatomically compartmentalised, forming defined cortical, and medullary regions each differentially supporting critical aspects of thymus‐dependent T‐cell maturation. Importantly, the specific functional properties of thymic cortical and medullary compartments are defined by highly specialised thymic epithelial cells (TEC). For example, in the medulla heterogenous medullary TEC (mTEC) contribute to the enforcement of central tolerance by supporting deletion of autoreactive T‐cell (...) clones, thereby counterbalancing the potential for random T‐cell receptor generation to contribute to autoimmune disease. Recent advances have further shed light on the pathways and mechanisms that control heterogeneous mTEC development and how differential mTEC functionality contributes to control self‐tolerant T‐cell development. Here we discuss recent findings in relation to mTEC development and highlight examples of how mTEC diversity contribute to thymus medulla function. (shrink)

A total of 944 expressed sequence tags generated 2212 EST loci mapped to homoeologous group 1 chromosomes in hexaploid wheat. EST deletion maps and the consensus map of group 1 chromosomes were constructed to show EST distribution. EST loci were unevenly distributed among chromosomes 1A, 1B, and ID with 660, 826, and 726, respectively. The number of EST loci was greater on the long arms than on the short arms for all three chromosomes. The distribution of ESTs along chromosome arms (...) was nonrandom with EST clusters occurring in the distal regions of short arms and middle regions of long arms. Duplications of group 1 ESTs in other homoeologous groups occurred at a rate of 35.5%. Seventy-five percent of wheat chromosome 1 ESTs had significant matches with rice sequences, where large regions of conservation occurred between wheat consensus chromosome 1 and rice chromosome 5 and between the proximal portion of the long arm of wheat consensus chromosome 1 and rice chromosome 10. Only 9.5% of group 1 ESTs showed significant matches to Arabidopsis genome sequences. The results presented are useful for gene mapping and evolutionary and comparative genomics of grasses. (shrink)

To localize wheat ESTs on chromosomes, 882 homoeologous group 6-specific ESTs were identified by physically mapping 7965 singletons from 37 cDNA libraries on 146 chromosome, arm, and sub-arm aneuploid and deletion stocks. The 882 ESTs were physically mapped to 25 regions flanked by 23 deletion breakpoints. Of the 5154 restriction fragments detected by 882 ESTs, 2043 were localized to group 6 chromosomes and 806 were mapped on other chromosome groups. The number of loci mapped was greatest on chromosome 6B and (...) least on 6D. The 264 ESTs that detected orthologous loci on all three homoeologs using one restriction enzyme were used to construct a consensus physical map. The physical distribution of ESTs was uneven on chromosomes with a tendency toward higher densities in the distal halves of chromosome arms. About 43% of the wheat group 6 ESTs identified rice homologs upon comparisons of genome sequences. Fifty-eight percent of these ESTs were present on rice chromosome 2 and the remaining were on other rice chromosomes. Even within the group 6 bins, rice chromosomal blocks identified by 1-6 wheat ESTs were homologous to up to 11 rice chromosomes. These rice-block contigs were used to resolve the order of wheat ESTs within each bin. (shrink)

This article presents an approach we have developed for the assessment of public research institutions in the natural sciences and engineering. The method consists in the exploitation of available S&T databases and indicators, including bibliometrics (databases of scientific as well as technical literature), patents, and internal data. Results are therefore derived through multiple databases. The objective is to profile a specific national R&D community in order to assess the positioning of a public institution or program within the community to which (...) it belongs. It involves obtaining the profile of the evolution of an R&D area, the R&D performers, the financing and support institutions, the users of R&D results, and the interactions between all these actors. Beck, 1997. 218 pp. Cloth, DM 48, SFr 44.50, ÖS 350.Looking at the dust cover of this volume, which features a Pompeiian wall painting with an impetuous satyr tightly grasping a nude, buxom, and resistant female, a reader might be led to expect a conventionally shallow literary biography of Ovid, the poet of erotic (...) enjoyment. Probably Holzberg had nothing to do with the choice of the cover, which was designed only to attract buyers. If he did, he would have been ironically aware of the surprises he had in store for unsuspecting readers: there is little in this book to excite the average modern satyr. Instead of titillating his audience with erotic images, Holzberg focuses our attention on the elegiac system, from which, as he shows, the poet Ovid ingeniously worked.The reader receives warning in the foreword, where Holzberg says little about the historical personage Ovid but directs us instead to the variety of roles the poet Ovid developed in his different works. It is these roles that have familiarized us with Ovid, not the historical facts that have accumulated around him [End Page 651] outside his poems. Holzberg declares, then, that he wishes to read Ovid’s poetry as “novels,” that is, as fictional accounts by a fictional speaker who uses the first person.Before proceeding to the poetic works in chronological order, Holzberg devotes two important introductory chapters to discussing his approach first to the work, then to the poet. That of course reverses the order of his subtitle, but by way of correction: this writer and this book are primarily interested in the poetry that Ovid created and that we can still critically read: the poet is far more problematical. So we start with a paradox: our present time is an Ovidian Age (aetas Ovidiana), as was the era in cultivated Western Europe about a thousand years ago. We probably read Ovid today as well as, if not better than, the people who rushed to multiply Ovidian manuscripts in that previous era. However, the question is: do we read and hear Ovid with the awareness of those original Roman audiences, nearly a millennium earlier, who enjoyed what we may consider the first aetas Ovidiana, when Ovid himself was present to register his amazing effects in Rome (and to recall them fondly in exile)? The answer that Holzberg produces is: no, we don’t read Ovid as a Latin audience; instead, we pick out what makes him like us, modern. To read his works as Holzberg wants, then, we must first grasp the features which belong to the doctus poeta. Ovid was what Holzberg calls “a poet between texts” (a clever variation on Fraenkel’s famous phrase for Ovid, “a poet between two worlds”). That stresses the literary practice of intertextuality rather than the cultural interrelationships that occupied Fraenkel and his contemporaries. It also justifies the role of Holzberg, who steps in to help the helpless “modern reader” and to explain some of the key intertexts that functioned directly for Ovid’s first audiences. Similarly, a modern reader needs to step back from the trap of the plausibly witty story that Ovid’s first-person speaker narrates, to appreciate the conventions with which the poet ably plays for the amusement of his well-read audiences; and this means understanding what Holzberg calls the “elegiac system,” which serves as the basis of all Ovid’s work. Recognizing the fundamental elements of the system, we can see better how he modifies and innovates with familiar material, whether it is a form like that of the exclusus amator or the interesting variations he creates on the “slavery of love.”Holzberg’s approach to Ovid the poet is deliberately skeptical. Our only contemporary “evidence” about him is the dubious and fictional series of details and events which emerge from the mouth of the “speaker” in the poems. Holzberg distrusts even the prodigious memory of Seneca the Elder. And it is notable that Tacitus, who had the opportunity of giving Ovid an obituary around A.D. 17, in what survives of the... La dernière édition des Lettres sur les animaux, ouvrage de l'encyclopédiste mineur Charles-George Le Roy, date de 1896. Cette nouvelle édition propose une présentation très respecteuse de la pensée originale de l'auteur, elle précise dans quelles circonstances les divers éléments du livre furent successivement publiés et retrace son évolution depuis les articles HOMME (Morale) et INSTINCT de l'Encyclopédie jusqu'à l'édition complète de 1802. L'introduction situe les Lettresdans l'uvre de Le Roy qui, comptant l'écriture parmi ses activités, fut d'autant plus mêlé (...) aux conflits d'idées de l'époque. Des documents inédits permettent d'établir avec exactitude combien Le Roy a su mettre à profit ses fonctions de lieutenant des chasses des Parcs de Versailles pour exercer ses talents d'auteur. A la lumière de divers autres documents, et parmi eux des inédits, il apparaît que Le Roy fréquentait quelques-uns des penseurs les plus connus de l'époque (Condillac, Buffon, Diderot, Helvétius, d'Holbach), ainsi que des personnalités de la haute société (en particulier Mme de Marchais), deux mondes don't l'influence est perceptible dans les Lettres sur les animaux. The field of cognitive modeling has progressed beyond modeling cognition in the context of simple laboratory tasks and begun to attack the problem of modeling it in more complex, realistic environments, such as those studied by researchers in the field of human factors. The problems that the cognitive modeling community is tackling focus on modeling certain problems of communication and control that arise when integrating with the external environment factors such as implicit and explicit knowledge, emotion, cognition, and the cognitive (...) system. These problems must be solved in order to produce integrated cognitive models of moderately complex tasks. Architectures of cognition in these tasks focus on the control of a central system, which includes control of the central processor itself, initiation of functional processes, such as visual search and memory retrieval, and harvesting the results of these functional processes. Because the control of the central system is conceptually different from the internal control required by individual functional processes, a complete architecture of cognition must incorporate two types of theories of control: Type 1 theories of the structure, functionality, and operation of the controller, and type 2 theories of the internal control of functional processes, including how and what they communicate to the controller. This book presents the current state of the art for both types of theories, as well as contrasts among current approaches to human-performance models. The emergence of the new coronavirus at the beginning of 2020, considered a public health emergency due to its high transmission rate and lack of specific treatment, led many countries to adhere to social isolation. Although necessary, social isolation causes important psychological changes, negatively affecting the health of the population, including the older population. The aim of this study is to propose a 4-week, home-based physical exercise protocol for older people in social isolation and evaluate whether will promote positive changes (...) in psychological variables such as anxiety, mood, depression, and stress, and in the variables sleep, quality of life, and physical capacities in the older adults. The sample will be selected in a probabilistic way from individuals aged 60 years or more from the city of Itajaí. Of these, half will perform a home-based resistance training protocol, with 3 weekly sessions, for 4 consecutive weeks. For group allocation, patients will be randomized with a computer-generated 1:1 allocation to the physical exercise group or control group. Outcomes will be depressive symptoms, sleep quality, quality of life, stress, mood states, anxiety, and functional capacity, evaluated at baseline, after 4 weeks, and after 15 days of follow-up. This study will offer a home-based exercise protocol for older adults, with load progression and remote monitoring, thus filling a gap in the provision of PE in this population. The results will be able to identify possible improvements not only in physical health, but also in quality of life and mental health.Clinical Trial Registration: The trial registration was carried out in the Brazil Clinical Trials Registry.. This study investigated the performance of boys with psychopathic tendencies and comparison boys, aged 9 to 17 years, on two tasks believed to be sensitive to amygdala and orbitofrontal cortex func- tioning. Fifty-one boys were divided into two groups according to the Psychopathy Screening Device (PSD, P. J. Frick & R. D. Hare, in press) and presented with two tasks. The tasks were the gambling task (A. Bechara, A. R. Damasio, H. Damasio, & S. W. Anderson, 1994) and the (...) Intradimensional/ Extradimensional (ID/ED) shift task (R. Dias, T. W. Robbins, & A. C. Roberts, 1996). The boys with psychopathic tendencies showed impaired performance on the gambling task. However, there were no group differences on the ID/ED task either for response reversal or extradimensional set shifting. The implications of these results for models of psychopathy are discussed. The papers in this volume bring together a number of scholars working on topics ranging from contemporary ethics to the history of philosophy. This wide scope reflects the comprehensive nature of Dietrich von Hildebrand's philosophic vision. His career includes foundational works on such subjects as ethics, philosophical method, aesthetics, and the nature of love. One struggles to imagine how Hildebrand managed to fit all this into a life that also includes extensive anti-Nazi political activism during World War II. Within this (...) broad scope of Hildebrand's work, one finds a number of important philosophical problems that beg for further development, such as his conception of personal existence, his rich and pluralistic conception of beauty, or his distinction between person and personality. In several places, however, the development of Hildebrand's thought must involve a correction or even an outright challenge to his reasoning. Each of these papers does just this, applying the insights of Hildebrand to a number of diverse philosophical problems, while also advancing and challenging his thought in various ways. Despite this wide diversity, three central issues appear again and again in these papers, and represent some of Hildebrand's most important contributions to philosophy: (i) his method of phenomenological realism, (ii) his conception of value and value-response, and (iii) his personalism. Digital personal data is increasingly framed as the basis of contemporary economies, representing an important new asset class. Control over these data assets seems to explain the emergence and dominance of so-called “Big Tech” firms, consisting of Apple, Microsoft, Amazon, Google/alphabet, and Facebook. These US-based firms are some of the largest in the world by market capitalization, a position that they retain despite growing policy and public condemnation—or “techlash”—of their market power based on their monopolistic control of personal data. We (...) analyse the transformation of personal data into an asset in order to explore how personal data is accounted for, governed, and valued by Big Tech firms and other political-economic actors. However, our findings show that Big Tech firms turn “users” and “user engagement” into assets through the performative measurement, governance, and valuation of user metrics, rather than extending ownership and control rights over personal data per se. We conceptualize this strategy as a form of “techcraft” to center attention on the means and mechanisms that Big Tech firms deploy to make users and user data measurable and legible as future revenue streams. Human neural and behavioral activities have been reported to exhibit fractal dynamics known as 1/f noise, which is more aptly named 1/f scaling. Some argue that 1/f scaling is a general and pervasive property of the dynamical substrate from which cognitive functions are formed. Others argue that it is an idiosyncratic property of domain‐specific processes. An experiment was conducted to investigate whether 1/f scaling pervades the intrinsic fluctuations of a spoken word. Ten participants each repeated the word bucket over 1,000 (...) times, and fluctuations in acoustic measurements across repetitions generally followed the 1/f scaling relation, including numerous parallel yet distinct series of 1/f fluctuations. On the basis of work showing that 1/f scaling is a universal earmark of metastability, it is proposed that the observed pervasiveness of 1/f fluctuations in speech reflects the fact that cognitive functions are formed as metastable patterns of activity in brain, body, and environment. In this paper, I argue for an extension of relational accounts of the imago Dei which includes a kind of priestly relation to the created order. In this relation, humanity is intended to ensure the independent flourishing of creation in a way reflective of the kind of communion we ought to have with one another. Through an analysis of the brokenness of these relationships, I argue human oppression of other humans and ravaging of creation are born of the same brokenness (...) in such a way that they contribute to one another as distortions of human teleological communion. By drawing on Social Identity Approach in group psychology, I can offer an account of shared human identity out of which humanity acts in distorted ways as a group. By describing oppression and ravaging in terms of broken communal telos and group action, I offer a way forward for relating humanity to the created order in a way that neither instrumentalizes creation, nor flattens the distinctiveness of human creation in the image of God.
